I built my first scooter with a 72v 3000w kit. Now wondering if I can also use my investment in lithium batteries in travel trailer I am building.

All else being equal, if I use a buck step down to 12v, do the amphours increase x6 ?

My scooter currently has a cheap 12v step down for a single headlight bulb & a battery display. Didn’t use any criteria except for that 72v to 12v “cheap”. If I can use the lithium batteries I already have for a travel trailer, what are the specs I should look for, and a possible source, for a quality, efficient 72v to 12v step down ?

You'll get a proportional increase in amp hours, minus conversion losses due to inefficiency. Voltage converters tend to be most efficient when loaded up, and inefficient at low loads. The cheap Chinese kind aren't reliable at any more than about half their amp rating, though.
